The Human Amyloid Beta 1-40 (High Sensitivity) ELISA Kit provides quantitative detection of human Aβ1-40 with an LOD of 1.46 pg/mL and a range of 4.69–300 pg/mL. Aβ1-40 is the most abundant amyloid isoform in plasma and CSF. Paired with ENZ-KIT273 (Aβ1-42), it enables calculation of the Aβ42/40 ratio — the clinically preferred metric for blood-based amyloid pathology screening.

Amyloid Beta 1-40 (Aβ40) is the predominant amyloid-β peptide produced through sequential cleavage of amyloid precursor protein (APP) by β-secretase and γ-secretase. Although Aβ40 is less aggregation-prone than Aβ42, it is an important component of amyloid-β biology and serves as a reference peptide for calculating the Aβ42/Aβ40 ratio. This ratio generally provides better discrimination of cerebral amyloid pathology than Aβ42 alone and correlates with amyloid PET status. Plasma Aβ42/Aβ40 thresholds are assay-dependent and require validation for the intended use. The ratio is an important biomarker in Alzheimer’s disease research and may support blood-based assessment of amyloid pathology when measured using sufficiently validated assays. Aβ40 is also studied in cerebral amyloid angiopathy and in research evaluating γ-secretase activity and modulation.
