The eukaryotic translation initiation factors (eIFs) regulate translation at the level of messenger RNA engagement by the ribosome. eIF-2alpha promotes the binding of the initiator tRNA to 40S ribosomal subunits, and is a key signaling component of the unfolded protein response (UPR). eIF-4E recruits mRNAs to the eIF4F complex through binding of their 5 N7-methylguanosine cap. eIF-4E binding proteins-1 and -2 are translational repressors, sequestering eIF-4E in conditions of low cellular demand for translation. Their dissociation from eIF-4E is regulated by phosphorylation in response to growth-promoting stimuli.
