The DEEPSEE® ENZ-GEN444-0020 is designed for detection of MDM2 gene amplification and copy number alterations of chromosome 12 by fluorescent in situ hybridization (FISH). The probe mix is suitable for identifying genetic abnormalities in cultured cells. DEEPSEE® ENZ-GEN444-0020 contains two fluorescently labeled probes. The MDM2 probe (orange) targets the MDM2 oncogene locus at 12q15, which is commonly amplified in a wide range of soft tissue sarcomas and other cancers. The centromere enumeration probe CEN 12 (green) targets the alpha satellite sequences within the centromere of chromosome 12, serving as an internal reference control for chromosome 12 copy number. Hybridization of a target DNA specimen with Enzo’s DEEPSEE® ENZ-GEN444-0020 produces bright and intense orange and green fluorescent signals, enabling direct visualization of MDM2 amplification and chromosome 12 copy number changes. The MDM2-specific FISH probe is optimized to detect the 12q15 locus on chromosome 12 in cultured cell lines from Homo sapiens The chromosome-specific centromere enumeration FISH probe is optimized to detect alpha satellite sequences within the centromere of chromosome 12 in cell lines from Homo sapiens CEN 12 hybridizes at 12p11.1–12q11.1 SEEBRIGHT® Mounting Medium with DAPI is an antifade reagent that minimizes photobleaching. Use of this mounting medium significantly extends the lifetime of fluorescent signals. The inclusion of DAPI allows for staining of the nucleus.

Fluorescence in situ hybridization (FISH) is a molecular cytogenetic technique that uses fluorescently labeled DNA probes to detect and localize specific DNA sequences on chromosomes. FISH is widely used in research and clinical laboratories for the detection of chromosomal copy number changes, gene amplifications, deletions, and structural rearrangements associated with cancer and genetic disorders. DEEPSEE® directly labeled FISH probes are supplied as pre-diluted hybridization solutions, enabling high-sensitivity detection without additional labeling steps. SEEBRIGHT® Mounting Medium with DAPI is recommended for use with this probe.
