W45 is a potent peptide-based, membrane-impermeant inhibitor of cyclic GMP-dependent protein kinase Iα and Iβ. It displays selectivity for cyclic GMP-dependent protein kinase Iα compared to cyclic AMP-dependent protein kinase with a ratio of approximately 600 fold (Dostmann et al. 2000).
Fusion of W45 to membrane translocation sequences from HIV-1 Tat protein or Drosophila Antennapedia homeodomain results in the membrane-permeant fusion peptides DT-2 (Prod. No. BLG-D051) and DT-3 (Prod. No. BLG-D052), respectively. Since the highly charged W45 cannot cross cell membranes it can be used as a control in applications of DT-2, DT-3 and the proteolytically stable D-amino acid congener of DT-2, (D)-DT-2 (Prod. No. BLG-D110).
