Spautin 1 inhibits USP10 and USP13 with IC50’s of 0.6 μM and 0.7 μM, respectively. These deubiquitylases target the Beclin-1 subunit and promote degradation of Vps34-PI3 kinase complexes. Its effect on autophagy is Beclin-independent. Spautin induces apoptosis of cancer cells under starvation conditions.
