pURE1

The pURE vector series are especially designed for generation of template DNA for in vitro transcription/translation kit, PURESYSTEM®. These vectors for transcription and translation are constructed from T7 promoter, and ribosome binding site (RBS) at a suitable distance upstream from the start codon ATG. Therefore, the 5′-end of gene of interest which includes the ATG start codon must be inserted into NdeI, NcoI or SphI site of pURE1, pURE2 or pURE3, respectively. The pURE4 is available for blunt 5′-end ligation with EcoRV site. It is also necessary to confirm the direction of the gene of interest if it is inserted into single restriction site.
