Human DNA fragmentation factor (DFF) is a heterodimeric protein complex consisting of the DNase inhibitor protein DFF45 and the caspase-activated nuclease DFF40 (CPAN). The mouse homologue is the caspase-activated DNase complex (CAD Complex) consisting of the caspase activated DNase CAD and its inhibitor ICAD. Upon cleavage of DFF45(ICAD), the caspase activated DNase DFF40(CAD), is released, becomes activated and eventually causes the degradation of DNA in the nuclei: the hallmark of apoptotic cell death. The human caspase activated DNase that is inhibited by DFF45 has been cloned.
