IU1 is a cell-permeable, reversible, and selective inhibitor of the proteasome-associated DUB USP14 (IC50 < 4 µM). IU1 enhances proteasome activity and substrate proteolysis. IU1 promotes cell survival under stress conditions and selectively stimulates ubiquitin-dependent protein degradation in vitro.
