Negative control for Fc chimeric proteins

Negative control for Fc chimeric proteins. Does not bind to a series of ligands in an ELISA assay. Does not inhibit binding between the thymoma cell line EL-4 expressing β3-integrin and astrocytes expressing high level of Thy-1 in mice.

Thy-1 is a glycosyl phosphatidylinositol (GPI)- anchored glycoprotein of the immunoglobulin superfamily expressed in various cell types, particularly in T-lymphocytes and cells of the neuronal system. Thy-1 is involved in cell adhesion, and the presence of a RLD motifs, which is recognized by intengrins, strengthen the hypothesis that the Thy-1:integrin interaction is responsible for cell attachment. In astrocytes, binding of Thy-1 to β3-integrin triggers tyrosine phosphorylation of focal adhesion proteins.

No. ALX-203-004): Negative binding to members in TNF-superfamily, including, but not limited to, CD40L, TRAIL, TNF, CD137L, FasL at various concentrations as indicated. (The CD40L and TRAIL used in the assay are fully capable to bind to their native biological receptors , i.e. CD40 and TRAIL-R2, respectively, as shown by the red square and diamond.)

Reconstitution	Reconstitute with 50µl sterile distilled water. Further dilutions should be made with medium containing 5% fetal calf serum or a carrier protein.
Source	Produced in HEK 293 cells. The extracellular domain of mutated hThy-1, hThy-1 (RLE) (aa 20-130), is fused to the Fc portion of human IgG1. The integrin binding sequence RLD of hThy-1 has been changed to RLE abolishing the binding of integrins.
Specificity	Does not bind to β3-integrin nor to any of a large number of tested TNF-superfamily members including rhsTRAIL, rhsCD40L, rhsAPRIL, etc.
