Baricitinib phosphate

Baricitinib is a selective JAK1 and JAK2 inhibitor. IC₅₀ values are 5.9nm for JAK1, and 5.7nM for JAK2. In cell based assays Baricitinib/INCB-28050 demonstrated inhibition of intracellular signaling of multiple pro-inflammatory cytokines including IL-6 and IL-23 at concentrations of < 50 nM, and also showed good efficacy in a number of animal models at doses of 10mg/kg and below. Baricitinib is in Phase III development as a potential treatment for rheumatoid arthritis. It is in Phase II development as a potential treatment for psoriasis and diabetic nephropathy.
