
BAD is the biologically active metabolite of benzamide riboside and a potent inhibitor of inosine 5′-monophosphate dehydrogenase (IMPDH). It was also found to be a potent and competitive inhibitor of human NAD kinase (Ki = 90 µM) and, at higher concentration and to a lower extent, of the bacterial enzyme. Furthermore, BAD is a non-hydrolysable substrate/inhibitor of NADases and PARP enzymes. BAD was used in co-crystallization experiments with NADases and PARP1.
