AS1842856 is a cell-permeable compound that specifically and potently inhibits the transcriptional activity of the Forkhead box O1 (Foxo1) protein (IC50 value of 33 nM). This inhibition is mediated by the direct binding of AS1842856 to the active form of Foxo1, but not to its Ser256-phosphorylated (inactive) form.

AS1842856 has been shown to preferentially inhibit Foxo1 over other related proteins such as Foxo3a and Foxo4. This selectivity makes it applicable for investigating the role of Foxo1 in various cellular processes, like metabolism, cell cycle regulation, and apoptosis. Moreover, AS1842856 can inhibit gluconeogenesis in liver cells and reduce glucose production in both in vitro and in vivo models, extending its use in the research of metabolic disorders, such as diabetes.
