14-3-3 proteins are dimeric phosphoserine/phosphothreonine binding proteins, which have been shown to be involved in the regulation of many crucial processes such as signal transduction, cell-cycle control, apoptosis, protein localization, phosphorylation-state and stability, transcription, metabolism, and malignant transformation. 14-3-3σ is the main 14-3-3 isoform that has been directly implicated in the development of cancer. The 14-3-3σ gene is directly regulated by p53 and is silenced by CpG methylation in a large number of carcinomas.
