We have not specifically tested metal chelators on the sirtuins. Reaction inhibition by EDTA or EGTA is possible although inhibition effect by these metal chelators and the concentration that would cause inhibition have not been determined by us. According to Chen L. et al. (Chen et al. (2010)), although zinc is a metal found to be critical in the zinc finger domain of sirtuins, exogenous zinc is found to be inhibitory. Activity of sirtuins can be rescued when hitting the reaction with EDTA, which binds up the extra zinc. The referenced data shows that EDTA did not inhibit SIRT1 under the specific reaction conditions described. However, different results may be observed if the concentration of chelators was to shift by an order of magnitude.
