Why is 4P curve fit curve recommended for ELISA data analysis and where can I find access to this data analysis software?

For the most accurate results, we normally recommend to use a 4-parameter logistic (4-PL) curve fitting software, as 4-PL fitting regression typically yields the most accurate standard curves for the determination of the analyte concentrations in unknown samples. Many plate readers do have the ability to perform a 4P curve fit analysis and the software is pre-loaded to the reader/computer. Check with plate reader manufacturer to confirm if this plate reader how to access and use this software. As an alternative, the data can be analysed in various statistics software programs, such as R/GNU-R or similar. It may also be done in MS Excel with the help of plugins that support 4- or 5-parameter logistic fit curves. Furthermore, some free online scripts for ELISA analysis are also available.
