In general, we would not advise holding the entire kit at 5-6 hours at ambient temperature if this can be avoided. Rather, we recommend returning each component to +4C as soon as possible after its use, and limiting the length of time that it is held at ambient temperature before its use. The unused plate strips (sealed back in the foil pouch with the desiccant) and standard should be returned to +4C after the first incubation step has started, the antibody returned to +4C after the second incubation step begins, and the conjugate returned to +4C after the 3rd incubation has begun.
