The SEEBRIGHT® fluorophore-labeled dUTPs are supplied as a 1 mM solution. We recommend to prepare working concentration of 0.3 mM by adding 10 µL of labeled dUTP to 23.3 µL of nuclease-free water. Store the diluted dUTP at -20°C after use. For all our fluorescent dUTPs except Green dUTP, the dTTP and dUTP ratio is 2: 1 (3.3µl dTTP and 1.7µl dUTP). The final concentration of labeled dUTP in the reaction tube is therefore 10.2 µM. For the Green 500 dUTP, the ratio dTTP and dUTP labelled is 1: 1 (2.5µl dTTP and 2.5µl dUTP). The final concentration of labeled dUTP in the reaction tube is therefore 15 µM. Finally, for the bio-16-dUTP or the digoxigenin dUTP, the ratio of dTTP and dUTP labeled is 1: 2 (1.7 µl dTTP and 3.3 µl dUTP). The final concentration of labeled dUTP in the reaction tube is therefore 19.8 µM.
