There are two main types of b-galactosidases in senescent cells: SA-b-galactosidase and lysosomal b-galactosidase. The latter can only be detected at pH 4.0. The cell pre-treatment solution is used to induce lysosomal alkalinization to minimize the lysosomal b-galactosidase activity.
