Z-VAD(OMe)-FMK (BML-P416) has methyl ester group making this irreversible caspase inhibitor cell permeable and therefore is used in cell based application.  Methyl ester makes this peptide cell-permeable and once inside the cell, the methyl ester is hydrolyzed to release the active free acid form.

Z-VAD-FMK (ALX-260-020) does not have a methyl ester group and is therefore cell impermeable. Z-VAD-FMK, the free acid form is suitable for cell free and in vitro assays
