Hoechst 33258 (ENZ-52402) is less cell permeable than other derivatives, however it also allows for quantitative measurements when plotted in a standard emission-to-content curve. Hoechst 33342 (ENZ-52401) is more readily cell-permeable as the a lipophilic ethyl group makes it more lipophiic. It is usually preferred for living cell staining. It may be used on live or fixed cells and is also applicable for cell cycle analysis and monitoring DNA condensation by flow cytometry. Exclusion of Hoechst 33342 dye is a characteristic common to stem cells, as well as chemotherapy-resistant cancer cells.
