What could be the reason for low response to Wnt induction with LEADING LIGHT® Wnt Reporter Assay, ENZ-61001?

If observing a low response to Wnt induction with LEADING LIGHT® Wnt Reporter Assay (ENZ-61001), please consider the following. The reporter cell line may lose its ability to respond to Wnt induction after reaching a high number of passages; you could therefore thaw cells with low
passage from liquid nitrogen. Cell density is in the assay plate could be too low.  Cells are not conditioned well due to incompletely replacing
growth medium with assay medium. Additional causes are listed in the troubleshooting guide at the end of the user manual.
