Nile Red, Sypro Orange and 1,8-ANS detect protein unfolding through dye interaction with newly exposed hydrophobic surfaces. They are sensitive to the hydrophobicity of the surrounding environment in which they are employed, and as such, these environmentally sensitive dyes tend to be less specific when detecting protein aggregation and are susceptible to hydrophobic compound interference.
