The ProteoStat® Thermal Shift Stability Assay kit (ENZ-51027) is used for direct monitoring of protein aggregation arising from a thermally-induced protein denaturation without exposing the hydrophobic regions through protein unfolding. The system minimizes problems normally encountered by environment-sensitive dyes, such as high background fluorescence from their interaction with detergents, membrane proteins or hydrophobic compounds. The kit is used for determining the temperature at which the bulk of the protein becomes aggregated under a given condition, which in turn is used to optimize conditions that enhance protein stability. Conditions that increase the aggregation temperature increase the stability of the protein. The kit is also used for determining the effect of buffers and other added excipients on overall stability of a protein, or to identify ligands or drugs that bind and confer structural stability to a protein of interest.
