The most significant advantage for Cyto-ID® Autophagy Detection kit is that it's a no-transfection assay. Transfections are transient and there is cost factor towards repurchase reagents for different experiments. The transfection approach can lead to false results arising from the spontaneous formation of GFP-LC3 aggregates in over-expressing cells. Transfections are generally less than 100% efficient, so some cells do not express the requisite GFP construct. The Cyto-ID® authophagy dye allows measurement of autophagy more consistently across the entire population of cells. The dye-based assay saves time while offering the flexibility to move from one cell line/type to another, without requiring genetic modification of each cell line. The dye is also suitable for labeling primary cells.
