The active PKA kinase provided in the kit is to be used as a positive control and is not intended for formation of a standard curve. The concentration of the PKA can vary between lots and the information about concentration is provided on the vial label. This concentration is then used to calculate a titration of this positive control. Performing the titration can tell you the point at which the active kinase is saturated. The control enzyme is used to show that the rest of the kit components are working and to help establish optimal parameters for the rest of the assay.
