No, the peptide substrate in the PKA kinase activity kit (ADI-EKS-390A) is not specific to PKA and therefore it can be phosphorylated by other serine/threonine kinases. This has to be taken into account if unpurified samples are used in the assay. For that reason, while crude lysate preparation will give results, it is difficult to be sure that the signal is only representing the activity of PKA. The solution for that is to purify samples as outlined in the kit insert. Alternatively, addition of kinase inhibitors that could specifically reduce the activity of other kinases can be added.
