The inhibitor is similar in properties to Probenecid. Cells lacking active MDR pumps do not require addition of efflux inhibitor. Depending on the cell line, Verapamil, MK-571 or Novobiocin, can be added to inhibit specific MDR pumps. Without inhibitors, MDR pumps can lower signal intensity and cause the dye to be pumped out of the cells rapidly following dye loading.
