Is charcoal-stripped FBS and serum free DMEM required for spike and recovery experiment with cell culture sample with Progesterone ELISA kit, ADI-900-011?

ANS: We suggest to prepare standard curve in the same unconditioned media used to culture samples.  General recommendation for diluting cell culture media is 1:10 and media used to make the curve would have to be at the same dilution with assay buffer.  End user is required to optimize specific dilution of the media to ensure matrix effects are eliminated with the appropriate dilution. The user manual contains Sample Handling recommendations and provides a protocol for the preparation of the standard

Progesterone concentrations were measured different samples including tissue culture media, human saliva, and serum. Progesterone was spiked into the undiluted samples of these media which were then diluted with the appropriate diluent and then assayed in the kit. Percent recovery and recommended starting dilutions are indicated in the user manual.
