it is easier to dilute samples than to concentrate them. In order to prepare cells for use with Calcineurin cellular activity assay kit (BML-AK816) we recommend initially using a minimal amount of lysis buffer (e.g. enough to cover the cells on the plate or a pellet for suspended cells). Typically this works out to be somewhere around 10 million cells per ml of lysis buffer but the cell type and how they are growing will influence this.
