If the test protein is a variant with near identical amino acid sequence with the exception of one residue, is one peak expected ?

There may be a possibility of different conformations of the same protein aggregating at different temperatures. The sample could have a mixture of full length protein and a fragment of the full length protein. So, one could expect different conformations of the protein that aggregate at different temperatures reflecting heterogeneity.  If the test protein has traces of BSA in the formulation, one would see more than one peak.
