I plan to use your ADI-900-020A kit to measure Endothelin-1 (ET-1) concentration in cell culture supernatant. Samples contain 10% fetal bovine serum (FBS). Will I need to extract them?

The ET-1 ELISA kit is suitable for use with tissue culture supernatants containing up to 10% FBS. In our spike and recovery assay we found that neither extraction nor dilution were necessary for this sample type. However, we strongly recommend that each user determines the optimal dilution for their particular samples. The basic principle of whether or not extraction will be necessary is whether or not ET-1 is present at a concentration in the sample that falls within the dynamic range of the assay following dilution of the sample to a point at which the matrix does not interfere with the detection of the analyte.
