The most common method is to cover tissue sections on glass slides in the hybridization solution with a glass cover slip in a moisture chamber, ideally sealed off with rubber glue solutions. An even distribution of the hybridization solution over the sample, and protecting the sample from drying out are vital for excellent, reliable ISH results. Covering in parafilm, cling film or similar are often done, but might lead to suboptimal results.
