The amount of tissue or number of cells to use is entirely dependent on the experimental conditions. It is recommended to start with approximately 2-5 million cells/ml of HCl for an initial test with one or two representative samples. Depending on the results of that test, future samples can be appropriately titer. Remember, it is easier to dilute a concentrated stock than to start the experiment all over again.
