The Protein A ELISA Kit (ADI-900-057) recognizes natural and recombinant forms of Protein A. Four Protein A constructs were evaluated (post boiling) in the assay: Natural Protein A from S. aureus (Millipore); Recombinant Protein A from E. coli (Repligen); C: Recombinant Cys-Protein A from E. coli (GE); Recombinant alkaline-resistant Protein A variant from E. coli (MabSelet SuRe from GE). Spike and recovery experiments of Protein A constructs were evaluated in the presence of Human IgG. Intra-assay precision was determined by assaying 16 replicates of three buffer controls containing Protein A in a single assay. Inter-assay precision was determined by measuring buffer controls of varying Protein A concentrations in multiple assays over several days. Sensitivity and linearity in two different buffers have been determined.
