M. Broillet, et al (2001) and J. Chatton, et al (2002) reported that light and divalent cations, such as Ca2+ or Mg2+ artificially enhanced the fluorescence intensities of NO as detected by DAF-2. However, N. Suzuki, et al (2002) demonstrated that these factors did not interfere with DAF-2 itself, but rather increased the rate of NO release from the NO donors S-nitrosocysteine, DETA NONOate and sodium nitroprusside used by Broillet et al, and Chatton, et al. Furthermore, N. Li, et al (2003) and H. Ichimura, et al (2003) reported that no light effects were found to influence the detection of NO by DAF-2. S. Brodsky, et al (2002) used an NOS inhibitor to also show that Ca2+ exerted no influence on DAF-2 detection of NO. It should be noted that excitation intensities and irradiation times should be selected carefully since DAF-2 and other such fluorescent probes may bleach over time.
