It is possible to kinetically monitor the ability of the cells to proliferate with CYTO-ID® Red long-term cell tracer kit (ENZ-51037) or CYTO-ID ®Green long-term cell tracer kit (ENZ-51036). Color choice depends upon the instrumentation available and whether or not the cells contain GFP. CYTO-ID®red long term cell tracer kit works well for GFP expressing cells. Cells would be pre-labeled with the dye and seeded onto the substrate. Cells could then be imaged or analyzed by FACS at various time points. Every time a cell divides, the fluorescent signal will decrease by half in the resulting daughter cells. During the 100 hour incubation period the cells actively divide. At least 4-5 population doublings occur in this period. As expected, after each population doubling, the fluorescence per cell decreases by half. Beyond 96-120 hours, the fluorescence is so low that we can no longer distinguish between the stained and unstained cell populations.
