The BIOARRAYTM Terminal labeling kit kit was optimized with a 30-mer at 1 µg or 100 pmol, but a longer nucleotide sequence can be labeled with this kit. This would require optimization by increasing the incubation time from 15 minutes up to 1 hour as mentioned in step 3 of the protocol. By running a gel of labeled and unlabeled 100-mer, it would be possible to notice a shift in the size due to the biotin-ddUMP incorporation into the labeled sample. A mixture of unlabeled and gel shifted bands would indicate an incomplete reaction and further optimization would be necessary.
