Our recommendation is to analyze ELISA reads by 4P curve fit. Many plate readers do have the ability to perform a 4P curve fit analysis and the software is pre-loaded to the reader/computer. Excel cannot do a 4P logistic curve fit as a default. Specific plugins that support 4- or 5-parameter logistic fit curves might be installed. If the only option is to use the default Excel version to plot the results, standard curve can be generated by plotting OD values (or %binding) on the y-axis and concentration of standards on the x-axis. Trend line can then be obtained. Use this standard curve to read off the estimated concentration values of the unknown samples by finding the point where the standard curve intersects with the sample’s value on the y-axis (of either OD, B0 or both*). Then read off the x-axis value for that point.
