Antibody Drug Conjugates (ADCs) are a class of targeted biopharmaceuticals that combine the specificity of monoclonal antibodies with the potency of cytotoxic drugs. They are designed to selectively deliver cytotoxic payloads to specific cells, typically cancer cells, while sparing healthy cells. ADCs consist of three main components:

  • Monoclonal Antibody: The monoclonal antibody is engineered to recognize and bind to specific antigens present on the surface of cancer cells. This antibody acts as a homing device, guiding the ADC to its target.
  • Linker: The linker serves as a bridge between the antibody and the cytotoxic drug. It is designed to be stable during circulation in the bloodstream but capable of releasing the cytotoxic drug once the ADC reaches its target cell.
  • Cytotoxic Payload: The cytotoxic drug, often a potent chemotherapy agent, is attached to the ADC via the linker. Once the ADC is internalized by the target cell, the cytotoxic payload is released, leading to cell death.

By selectively delivering the cytotoxic payload to cancer cells, ADCs aim to enhance the therapeutic index by minimizing damage to healthy tissues and reducing side effects associated with traditional chemotherapy. ADCs offer the potential for targeted and personalized treatment options in the field of oncology, with the ability to improve efficacy and minimize systemic toxicity.
